Activation of the cGAS-STING innate immune response drives excessive and toxic release of pro-inflammatory cytokines in neurodegenerative conditions such Alzheimer’s and Parkinson’s disease. We have identified and developed potent and selective CNS-penetrant cGAS inhibitors capable of reducing brain inflammation in mouse models. These compounds are being further optimized towards identification of a pre-clinical candidate molecule.
